Second, storage conditions:

Third, the product principle:

The ADV-HR (Virus Aiding Reagent) reagent enriches the virus on the cell surface by physical adsorption, thereby increasing the efficiency of virus infection on the cells and promoting infection.

Fifth, the experimental example :

(1) Direct addition method (applicable to cell types that are more susceptible to infection)

1. When carrying out virus infection, add ADV-HR reagent directly into the culture dish/well/bottle and mix well.

2. Subsequent operations depend on the type of virus and the needs of the experiment.

(B) incubation method (applicable to cell types that are more difficult to infect)

Take a 6-well plate as an example:

1. Establish a 100 μL infection system in a 1.5 mL EP tube as follows:

1ADV-HR XuL

2 virus YμL

3 cell suspension (100-XY) μL (containing 1.0 × 10 6 cells)

(The ADR-HR dosage X should refer to Appendix 2 and the pre-experimental results; the virus dosage Y should refer to the virus titer and the MOI required for the experiment.)

Mix gently with a pipette and incubate in a 37 ° C cell incubator for 0.5-1 hour.

3. The cells in the infection system were gently suspended by a pipette, and all transferred to a 6-well plate to which 2 mL of the culture solution was previously added, and cultured in a 37 ° C, 5% CO 2 incubator.

4. Subsequent operations depend on the type of virus and the needs of the experiment.

Six, pay special attention to :

1. High concentration of ADV-HR is cytotoxic, affecting cell status and infection efficiency (see Appendix 2 for details) . We recommend that you perform a pre-experiment of the ADV-HR concentration gradient in the cells of interest.

2. This reagent is only used in combination with our company's virus products. All the consequences arising from the use of other company products are irrelevant to our company. The final interpretation of this product belongs to our company.

Experimental scale: 96-well plate (constant to 100 μL)

ADV-HR concentration: 1mg/mL

Description:

1. The final concentration of ADV-HR has no effect on cell status between 0 and 1.0×10 -2 mg/mL;

2. The cell growth was slow when the final concentration of ADV-HR was 2.5×10 -2 mg/mL;

3. When the final concentration of ADV-HR was 5.0×10 -2 to 1.0×10 -1 mg/mL, the cells were obviously broken. When 2.5×10 -1 to 5.0×10 -1 mg/mL, the cells shrunk or even stopped growing.

Suggest:

1. Perform a concentration gradient pre-experiment in the target cells before using ADV-HR;

2. It is recommended that the concentration of ADV-HR should not exceed 2.5 × 10 -2 mg / mL.
