First, the experimental conditions

Cometro High Performance Liquid Chromatography System;

Cometro (United States Connaught) 6000 LDI Pump ;

Softa ( Eastofa , USA) ELSD (Evaporative Light Scattering Detector), DT-70 °C, SC-30 °C;

Scienhome (Tianjin Airlines) Kromasil NH 2   Column

Mobile phase: acetonitrile - water = 70-30 ;

Flow rate: 1.0 ml/min ;

The concentration of lactose standard was 0.5mg/mL , 1mg/mL , 1.5mg/mL ;

Whey powder sample configuration: Take 0.1g powder into a 100mL volumetric flask   , dilute to the mark with water.

The injection volume is 20ul .

It can be seen from the figure that the peak shape of the sample is good and the column efficiency is high. Softa (ESF) ELSD (evaporative light scattering detector) has good reproducibility (RSD=0.72%) , and the linear relationship is good ( R 2 =0.999) .
